Flow cytometry analysis of BrdU incorporation in actively cycling Ramos cells. For in vitro labeling of cells, 20 µL of a 1 mM BrdU/PBS solution was added to 2 mL cell suspension containing 2x10^6 Ramos cells and incubated for 2 hours at 37C. Formaldehyde fixed cells were washed and permeabilized. To expose DNA incorporated BrdU, cells were treated with DNase for 1 hour at 37C. Unconjugated MA3-071 has been successfully used in immunofluorescence, immunohistochemistry, and flow cytometry applications. DyLight 488 conjugated MA3-071-D488 has been successfully used in flow cytometry applications.
Target Information
Bromodeoxyuridine (5-bromo-2-deoxyuridine, BrdU) is a synthetic nucleoside that is an analogue of thymidine. BrdU is commonly used in the detection of proliferating cells in living tissues, and can be incorporated into the newly synthesized DNA of replicating cells (during the S phase of the cell cycle), substituting for thymidine during DNA replication. Antibodies specific for BrdU can then be used to detect the incorporated chemical, thus indicating cells that were actively replicating their DNA. Binding of the antibody requires denaturation of the DNA, usually by exposing the cells to acid or heat.
