Flow Cytometry PBMC: Add 10 µL of MAB/10^6 PBMC in 100 µL PBS. Mix gently and incubate for 15 minutes at 2 to 8C. Wash twice with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze.
Flow Cytometry Whole Blood: Add10 µL of MAB/100 µL of whole blood. Mix gently and incubate for 15 minutes at room temperature 20C. Lyse the whole blood. Wash once with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. CD4, a single chain transmembrane glycoprotein, is found on a T cell subset (helper/inducer) representing 45% of peripheral blood lymphocytes. It is also present on 80% of thymocytes and at a lower level on monocytes. It is involved in recognition of antigen presented along with MHC class II by APCs. It serves as receptor for HIV. The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. The CD8 antigen acts as a corepressor with the T-cell receptor on the T lymphocyte to recognize antigens displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The coreceptor functions as either a homodimer composed of two alpha chains, or as a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains.
