In direct ELISAs and Western blots, approximately 15% cross-reactivity with recombinant human (rh) CA1, rhCA2, and rhCA3 is observed and 5% cross-reactivity with rhCA4, rhCA8, rhCA9, rhCA10, rhCA12, and rhCA14 is observed.

Carbonic anhydrase XII (CA XII), encoded by the CA12 gene, is one of the enzyme isoforms in the carbonic anhydrase family that catalyzes the reversible hydration of carbon dioxide into bicarbonate and protons. CA XII is a transmembrane protein expressed in various tissues, including the kidneys, intestines, and lungs, playing crucial roles in maintaining acid-base balance, pH regulation, and electrolyte transport. Its activity is significant in organs where fluid secretion and absorption are essential processes, impacting functions such as respiration and renal bicarbonate reabsorption. CA XII is involved in physiological processes like cell proliferation and differentiation by modulating the extracellular and intracellular pH, influencing tumor microenvironment dynamics. Aberrant expression of CA XII has been noted in several cancers, including breast and colorectal cancers, where it supports tumor growth and metastasis by maintaining the acid-base homeostasis conducive to cancer cell survival. Due to its surface expression and role in maintaining pH balance, CA XII is a target of interest for therapeutic interventions aiming to disrupt tumor progression and enhance the efficacy of cancer treatments.
