Immunofluorescence analysis of EIF2-alpha was done on 70% confluent log phase A549 cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.25% Triton™ X-100 for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature. Target Information

EIF2a (Eukaryotic translation initiation factor 2A) is a heterotrimer composed of three subunits (alpha, beta, and gamma). This translation initiation factor drives binding of initiator methionyl-tRNA to the 40S ribosome in an AUG-dependent manner to form the 43S pre-initiation complex. The polypeptide can be phosphorylated by related protein kinases activated in response to stress. Phosphorylated EIF2A inhibits EIF2B activity and prevents guanine nucleotide exchange.

AHO1182 was used in Immunocytochemistry, Western Blot to present in this study indicate that luteolin is markedly effective in disrupting the pathological PACT-PKR interactions to protect DYT-PRKRA cells against apoptosis, thus suggesting a therapeutic option for using luteolin to treat DYT-PRKRA and possibly other diseases resulting from enhanced PACT-PKR interactions.
