Primary antibody: Anti-NCOA3 at 5 µg/mL for 1 h at RT. Secondary antibody: Peroxidase rabbit secondary antibody at 1:10,000 for 45 min at RT. Staining: NCOA-3 as precipitated red signal with hematoxylin purple nuclear counterstain.

            This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an internal region of human NCOA3 isoform a protein.

            Product Specific Information

            Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.

            This antibody reacts with several isoforms of human NCOA3 protein (a, b, d, e and f). A BLAST analysis was used to suggest cross-reactivity with NCOA3 from human and macaque sources based on a 100% homology with the immunizing sequence. Expect partial reactivity with NCOA3 from dog, bovine, mouse and rat sources based on partial sequence homology. Cross-reactivity with NCOA3 from other sources has not been determined.

            Target Information

            AIB1-AC3 recognizes AIB1, a member of the SRC-1 family of nuclear receptor coactivators with an apparent molecular weight of 160kDa. The AIB1 gene has been mapped to region 20q12 and has been shown to be amplified in breast tumors and pancreatic cancer cell lines. AIB1 interacts in a ligand-dependent fashion with the estrogen receptor to enhance estrogen-dependent transcriptional gene activation.
