            Target Information

            PPIP5K2, also known as inositol hexakisphosphate and diphosphoinositol pentakisphosphate kinase 2, is a gene located on chromosome 5 in humans. This gene encodes a kinase involved in the phosphorylation of inositol pyrophosphates, which are essential for cellular signaling and regulation processes. Specifically, PPIP5K2 phosphorylates InsP6 and 1-InsP7 at the 5 position, playing a significant role in both cellular proliferation and metastasis. Recent studies have shown that PPIP5K2 can facilitate the growth and spread of non-small lung cancer (NSCLC) and colorectal carcinoma. It promotes cellular proliferation and migration independent of its kinase activity, in part by modifying its interaction with proteins like 14-3-3, affecting its subcellular distribution. Structural analysis of PPIP5K2 reveals that it possesses unique kinetic properties distinct from other inositol kinase isoforms. Genetic variants, such as the Arg837His missense variant, can alter its phosphatase activity, potentially impacting its functionality in signaling pathways. PPIP5K2 is intricately involved in epithelial-mesenchymal transition (EMT) mechanisms, further influencing cancer cell metastasis. Understanding the biochemical pathways and genetic variations associated with PPIP5K2 is crucial in comprehending its role in disease pathogenesis and therapeutic targeting.
