Immunofluorescence analysis of PTP1B was performed using 70% confluent log phase MCF-7 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.
This antibody has specificity for Human PTP1B/PTPN1.
Target Information
PTP1B (PTPN1) is a tyrosine-protein phosphatase which acts as a regulator of endoplasmic reticulum unfolded protein response. It mediates dephosphorylation of EIF2AK3/PERK and in turn inactivates the protein kinase activity of EIF2AK3/PERK. PTP1B also plays a role in CKII- and p60c-src-induced signal transduction cascades. It regulates the EFNA5-EPHA3 signaling pathway which modulates cell reorganization and cell-cell repulsion. PTP1B also regulates the hepatocyte growth factor receptor signaling pathway through dephosphorylation of MET.
