Slit guidance ligand 3 (SLIT3) is a gene located on chromosome 5q35.1, encoding a member of the SLIT family of proteins that play key roles in axon guidance and neuronal migration during development. SLIT3 functions as a secreted protein, influencing the directional growth and pathfinding of axons by interacting with its receptor, Robo, on the surface of neurons. This interaction is critical for establishing neural network architecture and maintaining proper neural connectivity. Beyond its role in the nervous system, SLIT3 is involved in processes such as angiogenesis and cell migration, impacting tissue development and repair. Dysregulation of SLIT3 expression or function has been associated with developmental abnormalities and various diseases, including cancer, where it may influence tumor cell invasion and metastasis.
