The Human Trappin-2/PI3 ELISA quantitates Hu PI3 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu PI3.
Principle of the method
The Human PI3 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

Peptidase inhibitor 3 (PI3), also known as Elafin, is a gene located on chromosome 20q13.12, encoding a protein that functions as a serine protease inhibitor. Elafin is primarily expressed in epithelial tissues, including skin and mucosal surfaces, where it plays a protective role against excessive proteolytic activity by inhibiting enzymes like neutrophil elastase and proteinase 3. This activity is crucial for maintaining tissue homeostasis and preventing inflammatory damage. PI3 is regulated by pro-inflammatory cytokines and is involved in modulating inflammatory responses and tissue repair processes. Its expression is often elevated in conditions characterized by inflammation, such as chronic obstructive pulmonary disease (COPD), psoriasis, and other inflammatory diseases, reflecting its role in guarding against protease-induced tissue damage. Research into PI3 focuses on its potential therapeutic applications in inflammatory diseases and its utility as a biomarker for assessing inflammation-related pathologies.
