The mouse Interleukin-2 (IL-2) ELISA kit facilitates the accurate quantification of mouse IL-2 in various biological samples, including murine serum, plasma, or cell culture medium. This highly specific assay is designed to exclusively detect both endogenous and recombinant forms of mouse IL-2.
Principle of the method
The mouse IL-2 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) employs a meticulously paired set of antibodies to capture and measure the target analyte. The microplate wells are pre-coated with a target-specific antibody that allows for the binding of the analyte during the assay. Upon the addition of samples or standards to the wells, they specifically bind to the immobilized (capture) antibody. The formation of a sandwich complex is achieved by introducing a second (detector) antibody. Subsequently, a substrate solution is added, initiating a reaction with the enzyme-antibody-target complex and generating a measurable signal. The intensity of this signal is directly proportional to the concentration of the target present in the original specimen.
Interleukin 2 (IL-2) is an immuno-modulatory cytokine that is important for the proliferation of activated T cells, differentiation of B cells, natural killer cells, monocytes and macrophages. IL-2 signals through the IL-2 receptor (IL-2R), a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL-4) and interleukin 7 (IL-7). The expression of the IL-2 gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a gene similar to IL-2 in mice leads to an ulcerative colitis-like disease that suggests an essential role of this gene in the immune response to antigenic stimuli.
