The Mouse MIF (Macrophage Migration Inhibitory Factor) ELISA quantitates MIF in serum, plasma, and other biological fluids
The Mouse MIF (Macrophage Migration Inhibitory Factor) solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the binding of the second (detector) antibody to the target on a different epitope from the capture antibody. An antibody conjugated with enzyme binds the formed sandwich. After incubation and washing steps to rid the microplate of unbound substances, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

MIF (macrophage migration inhibitory factor) was one of the first cytokine activities to be discovered and was initially described as a T cell-derived factor that inhibits the random migration of macrophages (Weiser 1989). Recently, MIF was rediscovered as a pituitary hormone that act as the counterregulatory hormone for glucocorticoid action within the immune system (Bernhagen 1993, Mitchell 1995). MIF was released from macrophages and T cells in response to physiological concentrations of glucocorticoids. The secreted MIF counter-regulates the immunosuppressive effects of steroids on immune cell activation and cytokine production (Bucala 1998). MIF plays a critical role in the host control of inflammation and immunity.
