Real Time PCR versus Digital PCR (dPCR): two methods, five applications

During this training we presented the principles for real-time PCR (qPCR) and digital PCR (dPCR); the two main chemistries used to generate the fluorescent signal.

Through five examples of applications we compared the methods of qPCR and digital PCR:

  • Absolute Quantification (ex. pathogen quantification)
  • Relative Quantification (ex. expression of genes)
  • Copy Number Variation (CNV)
  • Single nucleotide polymorphism (SNP) genotyping
  • Rare somatic mutation detection

We answered the following questions:

  • Is Absolute Quantification by qPCR really absolute?
  • How does digital PCR differ from qPCR?
  • What are the advantages and the limitations of these 2 techniques?
  • Are they complementary?
