Extraordinary simplified

Keeping abreast of the ability to increase proteome coverage with advances in instrumentation and workflows is of great interest to proteomics investigators.

In this webcast, the speaker will present first impressions of the Thermo Scientific Orbitrap Exploris 480 mass spectrometer equipped with a Thermo Scientific FAIMS Pro interface for shotgun proteomics. The performance of the new instrument is evaluated in data-dependent acquisition (DDA) and data-independent acquisition (DIA)* modes with fast scanning HCD methods in combination with front-end Field Asymmetric Ion Mobility Spectrometry (FAIMS).

This combination enables significantly increased proteome coverage when using single CVs and short LC gradients when connected to the Evosep One HPLC system. Moreover, the robustness of the new system is demonstrated by continuously evaluating peptide and protein identifications over thousands of LC-MS/MS analyses. Finally, raw sensitivity is evaluated analyzing dilution series of a HeLa digest by DIA, DDA and DDA with FAIMS.

Prof. Jesper V. Olsen studied analytical chemistry and obtained his PhD in biochemistry and molecular biology at University of Southern Denmark. He was involved in developing high-resolution mass spectrometry-based proteomics. In 2012, he was promoted to vice director of Novo Nordisk Foundation Center for Protein Research at University of Copenhagen, and in 2014 full professor.
