The Ion 16S™ Metagenomics Kit is designed for rapid, comprehensive and broad-range analyses of mixed microbial populations using the Ion Torrent™ semiconducter sequencing workflow. The kit permits PCR amplification of hypervariable regions of the 16S rDNA gene from bacteria. The amplified fragments can then be sequenced using the Ion PGM™ Sequencing 400 Kit on the Ion PGM™ platform and analyzed using the Ion 16S™ metagenomics analyses module within the Ion Reporter™ software.

The kit includes 2 sets of primers that can be used to amplify the corresponding hypervariable regions of the 16S rDNA gene in bacteria:
These comprehensive primer sets allow for accurate detection and identification of a broad range of bacteria down to the genus or species level. The primer sets are paired with Environmental Master Mix v2.0 that is optimized to tolerate high levels of PCR inhibitors and amplify targets from complex samples such as environmental, food, tissue and other challenging samples. Additionally, the kit includes a positive control for troubleshooting experiments.
Flexible
The primer sets are provided as two pools. Each pool can be used individually or in combination to allow for flexibility in experimental set up. When combined with the Ion Xpress™ Barcode Adapters kits, multiple polymicrobial samples can be combined in a single sequencing workflow.
Complete Workflow
Sequences generated using the Ion 16S™ Metagenomics Kit can be directly analyzed using the Ion 16S™ metagenomics analyses module within the Ion Reporter™ software enabling a rapid and semi-quantitative assessment of complex microbial samples.
Note: This kit is not compatible with the Ion Library Quantitation Kit (Cat. No. 4468802). For accurate quantitation of amplicons generated using this kit, use the Ion Universal Library Quantitation Kit (Cat. No. A26217).

For each sample, the protocol involves two PCR reactions, one with primer set V2-4-8 and one with primer set V3-6.7. However, the kit contains sufficient master mix for 100 reactions and each of the two primer pools has sufficient volume for 100 reactions. Thus, the kit contains sufficient reagents for 100 reactions/50 samples.
Note: 100 reactions is dependent upon on the master mix, which is the limiting reagent. As far as primers are concerned, there is sufficient volume to process another 50 samples.
