E-Gel CloneWell II Agarose Gels with SYBR Safe enable faster and more effective DNA fragment isolation for cloning workflows. The use of in-gel SYBR Safe stain and a blue-light transilluminator minimizes DNA damage and improves cloning efficiency compared to conventional methods. The E-Gel CloneWell II agarose gels are designed for use with the E-Gel Power Snap Electrophoresis Device (Cat. No. G8100).

E-Gel CloneWell II agarose gels enable you to:
Save time—no additional gel purification steps required after electrophoresis
Simplify DNA recovery—retrieve purified DNA directly from the recovery well with a pipette
Improve cloning results—minimize DNA damage and get more colony forming units for any cloning method

Fast DNA sample isolation in three steps
DNA fragment isolation with E-Gel CloneWell II precast gels is a simple 3-step procedure that eliminates conventional gel purification. To extract the DNA band of interest, load the sample into the loading wells, run the gel until the band reaches the recovery wells, and retrieve the sample. The covered sample is ready for cloning and does not require re-purification. Multiple bands from the one loading well can be collected during the same gel run.

DNA extracted from CloneWell II gels is compatible with common cloning methods such as Anza restriction cloning, TOPO cloning, and Gateway cloning.

Real time control of the sample
SYBR Safe DNA Gel Stain is incorporated into the gel itself, allowing you to visually monitor DNA band migration progress on the blue-light transilluminator of the E-Gel Power Snap Electrophoresis Device. The gel has reference marks for better control of the sample prior to the sample recovery step. If the target DNA passes the recovery well, the Reverse gel protocol allows for re-capture of the DNA band.

Improved cloning efficiencies
Even a short exposure of your DNA sample to UV light during visualization may lead to DNA damage and reduced cloning efficiencies. Using E-Gel CloneWell II gels together with the blue-light transilluminator minimizes UV damage and improves cloning efficiency when compared to conventional methods.

Safer alternative to ethidium bromide
The E-Gel CloneWell II agarose gel is pre-stained with SYBR Safe DNA stain that minimizes environmental impact and is safer than the ethidium bromide used in conventional preparative gel electrophoresis.

How can I view DNA on my E-Gel agarose gel with SYBR Safe DNA stain?

SYBR Safe-stained gels can be visualized with a standard UV transilluminator, a laser-based scanner equipped with an excitation source in the UV range or between 470 and 530 nm, or a blue-light transilluminator.

I accidentally allowed my DNA to run past the collection wells for my E-Gel SizeSelect II or E-Gel CloneWell II agarose gels. What should I do?

You can use the Reverse E-Gel Program to run the band back into the collection gel.

My E-Gel agarose gel has speckles when viewed in the imager. What should I do?

Here are some suggestions:

- Try cleaning the cassettes with alcohol and Kimwipes wipers.
- Try cleaning the camera lens.
- Try to adjust the exposure time and brightness options of the documentation system you are using.

My sample is leaking from the wells when running my E-Gel agarose gels. What happened?

Please ensure that you have not overloaded the well and that the wells were not damaged during comb removal.

I accidentally stored my E-Gel agarose gels at 4 degrees C instead of room temperature. Can I still use them?

While we recommend storage at room temperature, these gels will still be usable. Bring the gels to room temperature prior to the run for optimal conditions.
