General Description
• Oligomycin is a macrolide produced by Streptomyces diastatochromogenes that is toxic to other organisms
• Oligomycin blocks oxidative phosphorylation through inhibition of ATP synthase. The compound binds to the proton channel of ATP synthase and blocks mitochondrial proton import, resulting in accumulation of cytoplasmic protons
• Oligomycin also inhibits Na+/K+ exchange through Na+ occlusion within the Na/K-ATPase
• Oligomycin is provided as a mixture of A, B, and C isomers
Application
• Oligomycin inhibits respiration in mitochondria. It also inhibits ATP synthase
• Oligomycin can induce apoptosis in cultured human hepatocarcinoma and other mammalian cells in laboratory settings
