EpiLife is a sterile, liquid medium prepared with 60 μM calcium chloride for the long-term, serum-free culture of human epidermal keratinocytes and human corneal epithelial cells. This basal medium requires the addition of an appropriate growth supplement prior to use. EpiLife Medium greatly extends the in vitro lifespan of human epidermal keratinocytes compared to other serum-free media systems. This basal media is animal origin-free and contains essential and non-essential amino acids, vitamins, other organic compounds, trace minerals, and inorganic salts. It does not contain antibiotics, antimycotics, hormones, growth factors, or proteins. EpiLife Medium is HEPES and bicarbonate buffered and designed for use in an incubator with an atmosphere of 5% CO2 and 95% air.
To support plating and long term proliferation of human keratinocytes, EpiLife Medium must be supplemented with selected hormones and growth factors. We offer a choice of growth supplements for culturing human keratinocytes. Please refer to the EpiLife Medium user guide to determine the best supplement choice for your purposes. The supplements contain all of the growth factors, hormones, and extracts necessary for growth of human keratinocytes in EpiLife Medium.

Are the Keratinocyte SFM basal medium and supplement kit available separately?
The basal medium is not available for purchase individually, and can only be ordered as part of the Keratinocyte SFM kit (catalog # 17005-042). The supplements are available separately: Bovine Pituitary Extract is catalog # 13028-014 and the EGF is catalog # PHG0311.
Which serum-free media can be used to culture mouse keratinocytes?
The Keratinocyte SFM without calcium chloride (Cat. No. 37010-022) is appropriate for the culture of mouse keratinocytes. The medium should be supplemented with calcium chloride at a final concentration of 0.02 mM.
Which serum-free media can be used to culture human keratinocytes?
There are a variety of options for serum-free culture of human keratinocytes. Keratinocyte SFM (Cat. No. 17005-042) is a widely used kit that includes the basal medium, bovine pituitary extract (BPE) and Epidermal Growth Factor (EGF). If you would like a serum-free, defined culture system, the options are the Defined Keratinocyte SFM kit (Cat. No. 10744-019) or Epilife medium (Cat. No. M-EPI-500-CA) supplemented with EDGS (Cat. No. S-012-5). If you would like an animal origin free defined system, Epilife medium (Cat. No. M-EPI-500-CA) with S7 supplement (Cat. No. S-017-5) is an excellent choice.
