UVRAG is predicted to contain a coiled-coil structure and is likely to be metabolically unstable based on the occurrence of a strong PEST domain. It interacts with Beclin 1, leading to activation of autophagy and thereof inhibition of tumorigenicity of human colon cancer cells. The gene for UVRAG protein is localized on human chromosome 11q13. 5 and complements the ultraviolet sensitivity of xeroderma pigmentosum group C cells. The gene has been extremely well conserved during evolution, implying an important role for this gene product in cell metabolism. Chromosomal aberrations involving this gene are associated with left-right axis malformation, and mutations in this gene have been associated with colon cancer. The protein is highly expressed in human brain, lung, kidney and liver.
