How can I create a new variant call group in the HRM app on Thermo Fisher Connect, select a color for the variant, and assign wells to that group?
To create a new variant call group in the HRM app on Thermo Fisher Connect, follow these steps:
- Select the 'Quality Control & Results' tab.
- In the 'Quality Control & Results' pane, select your experiment of interest.
- Select the 'Derivative melt plot'.
- Click 'Actions', and then select 'Manual Call'.
- From the 'Manual Call' dialog box, you have two options:
- To assign the sample to an existing variant call: click 'Select Existing', choose the appropriate call from the 'Group' drop-down menu, and then click 'OK'.
- To create a new variant call: click 'Create New', enter a name for the new call in the 'Group' field, select a color, and then click 'OK'.
- Repeat the above steps to assign more manual calls.
- Click 'Analyze' to reanalyze the data using the manual calls.

Can I automate the extraction of files from Thermo Fisher Connect to any other cloud environment e.g., AWS, Egnyte etc?
It is not possible to automate extraction of files from Thermo Fisher Connect to any other cloud environment. However, if you are looking to automate data transfer, you do not need to go through Cloud.
- For newer instruments like Pro systems, you can transfer the data through an auto export directly from the instrument. During setting up the run, you can set up the auto export to the designated folder for LIMS pick up.
- For older instruments like Flex systems, you can do the same via the desktop software (auto export).

I have uploaded the .fsa files (fragment analysis data files) into Microsatellite Analysis (MSA) app on Thermo Fisher Connect (Cat. No A26811). I can no longer find the original .fsa files. Can I download them from the MSA app?
Yes. You may export .fsa files from the MSA software. This is done on the Setup page. From the Sample Table, select the samples to export. Then go to Actions and click on Export selected .fsa files. The files will download as a .zip file which needs to be extracted.

Is there a way to automatically upload my data into the Thermo Fisher Cloud app directly from the Applied Biosystems 3500 Genetic Analyzer, instead of using Data Collection Software?
If you are using the most recent version of Data Collection Software (v3.3), there is a way to automatically upload your data into the Thermo Fisher Cloud app from the instrument. When a run is complete, the data is stored in the results group folder that is specified in the plate record. The data will be automatically uploaded to your account if your User ID is the active ID. Results are saved in your account folders named by the plate and injections, not in the results group folder. Please go to page 43 of the 3500/3500xL Genetic Analyzer User Guide for Data Collection Software v3.3 (https://assets.thermofisher.com/TFS-Assets/LSG/manuals/100079380_3500v3_3RUO_UG.pdf) for details regarding the Thermo Fisher Cloud app.
If you are using Data Collection Software v3.1, you would have to manually upload your data into the Thermo Fisher Cloud app from your data folder on the computer.
