CaptureSelect FcXP Affinity Matrix is the next-generation CaptureSelect FcXL matrix, specifically designed for higher binding capacity. The selectivity remains identical; the ligand recognizes the CH3 domain of all human IgG subclasses (IgG1, 2, 3, and 4), while maintaining the elution characteristics of CaptureSelect FcXL Affinity Matrix, but the binding capacity is increased (>40 g IgG/L).

Features of this affinity matrix include:

  • Mild elution, making it suitable for Fc fusion proteins
  • Human-specific, no binding to bovine antibodies
  • Excellent scalability
  • Non-animal derived

CaptureSelect FcXP Affinity Matrix can be used for the purification of IgG and Fc fusion proteins directly from complex source materials and enables a robust and efficient purification process with excellent purity obtained in one step.

Free of animal components

CaptureSelect products contain affinity ligands based on recombinant single-domain antibody fragments (VHH) created by a proprietary technology. The VHH affinity ligand is a 12 to 15 kDa fragment comprising the three complementarity-determining regions (CDRs) that form the antigen-binding domain, efficiently produced in the yeast Saccharomyces cerevisiae by a production process free of any animal components (Animal Origin-Free).

Main characteristics:

  • Matrix: agarose-based, epoxide activated
  • Average particle size: 65 ± 10 μm
  • Ligand: CaptureSelect FcXP affinity ligand
  • Ligand coupling method: epoxide coupling
  • Binding capacity: 40–50 g IgG/L resin depending on flow rate, column height, and contact time
  • Elution conditions: 20 mM citric acid or acetic acid, pH 3–4, or 100 mM Tris, 2.0 M MgCl2, 40% (v/v) propylene glycol, pH 7
  • Flow characteristics: 50–200 cm/h (up to 2 bar)
  • Formulation buffer: 20% (v/v) ethanol
