Increased dynamic range detection of the FluxOR II Green Potassium Ion Channel Assay

CHO cells expressing hERG potassium channels were preloaded with the indicated reagent using the supplier protocol provided and stimulated with a solution containing 2mM thallium and 10 mM potassium to activate voltage gated potassium channels. The cells were exposed to different concentrations of the potassium channel blocker E-4031 and then detected using a Hamamatsu FDSS 6000 instrument. The FluxOR II Green Potassium Ion Channel Assay displayed a larger dynamic range of detection compared to the first-generation FluxOR assay or the Molecular Devices FLIPR™ assay, facilitating the analysis of a wider range of potassium channels, including low-expressed or weakly conducting channels.
