The Thermo Scientific Krios 5 Cryo-TEM is equipped with the Thermo Scientific Falcon 4i Direct Electron Detector, which can be combined with the innovative Thermo Scientific Selectris X Imaging Filter to greatly increase productivity and performance. The addition of our proprietary cold field emission gun (E-CFEG) can push performance even further to enable atomic-resolution imaging. (The E-CFEG, Selectris X Imaging Filters, and Falcon 4i Detector are also available as upgrade options for previous generations of Krios Cryo-TEMs.)
To meet the growing need for high-resolution, high-quality imaging, the Krios 5 Cryo-TEM can be equipped with a novel cold field-emission gun optimized for low energy spread (≤0.3 eV), which maximizes contrast at high spatial frequencies and makes it possible to efficiently solve structures at resolutions of 2.0 Å or less. For example, at a target resolution of 1.5 Å, contrast is enhanced 2x compared to our high-brightness X-FEG. Additionally, the E-CFEG enhances data throughput, thereby increasing productivity.
The post-column Selectris and Selectris X Imaging Filters improve the contrast of TEM images, resulting in high-resolution structures down to atomic resolution. The zero-loss filtering of the Selectris Filters removes noise caused by inelastically scattered electrons, producing an increased signal-to-noise ratio and better contrast. Designed for high stability, the zero-loss peak position of the Selectris Filters has minimized sensitivity to temperature variations in the environment, eliminating the need for frequent tuning.
With the Selectris Filters, zero-loss energy filtering is straightforward due to a thorough integration of software and hardware, along with extensive automation and exceptional stability. Every mechanical and electron-optical element has been designed for stability and data reproducibility, enabling the unattended and reliable acquisition of large datasets with narrow energy slit widths (<10 eV). The capability to use these narrow slit widths provides an additional boost in contrast for single particle analysis, enhancing signal strength, resolution, and throughput. To further increase the productivity and ease-of-use of cryo-EM workflows, Selectris Filters are fully integrated into Thermo Scientific Smart EPU Software, Thermo Scientific Tomography Software, and the instrument operation software.
A narrow energy slit width also means that less particles are needed to obtain an equivalent resolution as obtained with a wider energy slit (e.g., 370k vs 1M, as shown in the figure on the right). Consequently, this means that a Selectris X Energy Filter allows you to either obtain results faster with less data, or to attain higher resolution results using the same amount of data.
The Falcon 4i Direct Electron Detector is an ideal solution for both single particle analysis and cryo-electron tomography. It offers increased productivity and performance over previous Falcon Detectors with its unique combination of high image quality, high throughput, efficient lossless data compression (enabled via proprietary electron-event representation, EER), streamlined data management, and quality monitoring. Paired with the Krios 5 Cryo TEM, the Thermo Scientific Falcon 4i Direct Electron Detector produces exceptional quality data at high collection speeds and offers performance beyond the Nyquist frequency via sub-pixel precision counting.
Designed to protect cryo-electron tomography samples, the Thermo Scientific Vacuum Capsule prevents ice contamination, allowing lamellae created with cryo-focused ion beam milling to be transferred to a cryo-TEM without surface contamination. Compatible with Autoloader systems, the Vacuum Capsule facilitates easy and safe sample transfer, securely containing biological materials, including those that require specific biosafety levels.
The Krios 5 Cryo-TEM can be equipped with a 60°C heat decontamination solution, allowing it to be installed in facilities with certain biosafety-level containment requirements (e.g., BSL-3). This accessory enables pathogen research and comprehensive instrument service in these secure environments and can be combined with remote diagnostics and repair capabilities to accelerate instrument support and reduce the need for physical access.
Used to heat the interior of the microscope enclosure, the decontamination solution consists of both hardware and software. This includes heaters and temperature sensors within the enclosure, as well as additional components that prevent aerosol release. There is also a secondary workstation that provides microscope control from outside the containment area.
Software is used to initiate the heating cycle and define the duration of the process. It is also used to monitor heating using the multiple sensors within the enclosure.
The Krios 5 Cryo-TEM comes with a complete suite of automation software, including data management software that enables user-friendly single particle analysis data acquisition, Smart EPU Software that offers guided day-to-day operation, and a traffic-light UI element that clearly indicates the microscope’s status. Additionally, pre-defined workflow templates allow you to collect high-resolution data with only a few clicks.

Its modular organization provides a single convenient location to track experiments through individual steps in SPA and cryo-ET workflows.

Its new user-friendly interface enables both experienced and new users to jumpstart their tomography data acquisition.
Tomography 5 Software also connects with Thermo Scientific Tomo Live Software for automatic on-the-fly data reconstruction and review in any browser.
Tomo Live Software is an optional add-on for Tomography 5 Software that offers on-the-fly data monitoring through real-time reconstruction of tilt series into 3D volumes. It can automatically perform motion correction of fraction movies, fiducial-free tilt series alignment, and 3D reconstruction during tilt series recording. These reconstructed tomograms allow for impromptu data quality evaluation, identification of high- and low-quality tilt series, and exporting of selected tilt series and reconstructed tomograms for further processing, archiving, and sharing.
Smart EPU Software, an AI-enabled software solution, can analyze intermediate results, providing instant feedback, and steering data collection on the fly. The AI algorithms are based on years of cryo-EM knowledge, designed to replace decisions that experts need to make upfront and ensure that your instrument is working at optimal conditions, allowing you to focus on the science rather than on fine-tuning the microscope.
