TaqMan single-tube gene expression assays utilize TaqMan MGB probes, which include a reporter dye (such as FAM) at the 5' end and a non-fluorescent quencher (NFQ) at the 3' end. These probes are designed to increase the melting temperature (Tm) without increasing the probe length, allowing for the design of shorter probes and improving assay specificity.
