Gibco Viral Production Cells are a derivative of the HEK 293F cell line and are a core component of the LV-MAX Lentiviral Production System (A35684). Viral Production Cells have been adapted to a special chemically-defined, serum-free and protein-free LV-MAX Production Medium (A3583401). They are maintained in suspension culture and will grow to high density in LV-MAX Production Medium. Viral Production Cells are highly transfectable by LV-MAX transfection reagent (A35348) and generate superior lentiviral production compared to polyethyleneimine (PEI)-based lentiviral production methods.

Frozen cells are supplied in a vial containing 1 mL of cells at 1 x 107 viable cells/mL in LV-MAX Production Medium and 10% DMSO. The cells should be thawed directly into LV-MAX Production Medium.

The cells are typically grown in flasks on a shaker platform in a humidified, CO2 cell culture incubator. However, cultures are scalable in volume from 96-deep well blocks to 2-L flasks and bioreactors. Lentiviral vector harvest is typically performed at two days post-transfection.

Can I use Viral Production Cells (Cat. No. A35347, A35827) with the AAV MAX Helper-Free AAV Production System?

The Viral Production Cells 2.0 are specifically designed to work with the other components of the AAV MAX Helper-Free AAV Production System to provide maximal adeno-associated virus (AAV) titers. Viral Production Cells could be used but we don't recommend doing this as it will reduce system performance.

What are the major differences between LV-MAX and CTS LV- MAX products?

The LV-MAX and CTS LV- MAX products have equivalent performance. In contrast to the research use only LV-MAX products, CTS LV-MAX products have an additional intended use for cell and gene therapy applications, and are complemented with a more robust regulatory support package to enable clinical therapeutic applications. This means that to the best of our ability, we keep our CTS product specifications and release requirements up-to-date in accordance with the latest regulatory guidance specific for this intended use.

With the LV-MAX Production System, are the lentiviral particles stable in packaging media?

Lentiviral particles packaged in our LV-MAX Production Medium will be stable at 4 degrees C for 24 hours and will then begin to lose activity. For long-term storage, we recommend storing at –80 degrees C.
