LB agar is a popular, nutrient-rich medium made from casein peptone, yeast extract, sodium chloride, and agar. Casein peptone or tryptone is an enzymatic hydrolysate of casein. It supplies nitrogen, carbon, and amino acids. Yeast extract provides trace elements, vitamins (including B group vitamins), amino acids, and peptides. Sodium chloride supplies sodium cations for membrane transport and maintains osmotic balance. Agar is a solidifying agent of the growth medium.

One Shot LB Agar Plate composition is based on the Jeffrey H. Miller formulation and contains twice as much sodium chloride as the Lennox formulation. The inclusion of autoclaved glass beads allows for simple plate inoculation without the need for additional loops or spreaders.

As E. coli is unable to digest agar but can absorb nutrition from the LB medium, single colonies can be easily isolated and selected. On plates with a selective antibiotic only transformed E. coli with resistance can grow.

10 g/L casein peptone, 5 g/L yeast extract, 10 g/L NaCl, 15 g/L agar
