Ask our AI about this Product

Roswell Park Memorial Institute (RPMI) 1640 medium was originally developed to culture human leukemic cells in suspension and as a monolayer. RPMI 1640 has since been found suitable for a variety of mammalian cells including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes and carcinomas. Life Technologies offers a variety of Gibco™ RPMI 1640 modifications for a range of cell culture applications. Find the right formulation using the media selector tool.

This RPMI is modified as follows:
WithWithout
• L-glutamine• Sodium Bicarbonate
• Phenol Red
• HEPES

The complete formulation is available.

Gibco™ RPMI 1640 is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 contains biotin, vitamin B12 and PABA which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations.

Product Intended Use
For in vitro diagnostic use. CAUTION: Not for human or animal therapeutic use. Uses other than the intended use may be a violation of local law. Customers using Gibco™ RPMI 1640 in a manufacturing process, who have a submission with the FDA, may request a letter of authorization from Life Technologies to reference our Type II Drug Master File (DMF).

RPMI 1640 contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 uses a sodium bicarbonate buffer system (2.0 g/L) and therefore requires a 5-10% CO2 environment to maintain physiological pH.

Powder forms of Gibco™ cell culture medium require sodium bicarbonate supplementation, pH adjustment, and filtration at the time of preparation (see protocol for details).

For Research Use Only. Not for use in diagnostic procedures. Not for human or animal therapeutic use. Uses other than the intended use may be a violation of local law. Customers using Gibco RPMI 1640 in a manufacturing process, who have a submission with the FDA, may request a letter of authorization from Life Technologies to reference their Type II Drug Master File (DMF). For in vitro diagnostic use.

Frequently asked questions (FAQs)

How light sensitive is RPMI 1640 media? Should I also be protecting it from LED light?

While we know that different wavelengths of light are worse than others for exposure, we would recommend as a best practice to protect the medium from all forms of light exposure including LEDs, as much as possible to ensure optimal performance, as several components within the medium are light sensitive, such as vitamins.

Find additional tips, troubleshooting help, and resources within our Cell Culture Support Center.

What is the density (g/L) for RPMI 1640 Medium?

We have specific gravity information for RPMI 1640 Medium: 1.006 kg/L. In this case, the specific gravity is the same as density as the solvent is water.

What is the shelf life of my powdered media once I reconstitute it?

Expiration date of most Gibco reconstituted dry format media (AGT or DPM) has not been established; end users should assess performance and stability of this reconstituted media in a system that is relevant to their process. A dry format product (either DPM or AGT) will age upon storage and while we may not be able to detect which component(s) degrade since we can't measure every component, that doesn't mean that the reconstituted liquid will have the same stability as a fresh liquid made by direct weigh methods.

How long can I keep my media after supplementing with serum?

Generally speaking, media can be used for up to three weeks after supplementation with serum. There are no formal studies to support this, but it is the rule of thumb used by our scientists.

My medium was shipped at room temperature but it is supposed to be stored refrigerated. Is it okay?

We routinely ship media that require long-term storage in the refrigerator at room temperature. We have done studies on representative media formulations to show that media can be at room temperature for up to a week without a problem.

'The exposure of mammalian cells to UV irradiation induces the expression of immediate early genes such as c-jun and c-fos and activates the transcription factors AP-1 and NF-kappaB. JunD is one of the three members of the Jun family and shares some functional characteristics with c-Jun. In the present study, ... More
Role for 18:1 lysophosphatidic acid as an autocrine mediator in prostate cancer cells.

Journal:J Biol Chem
PubMed ID:12084719
Lysophosphatidic acid (LPA) is a lipid mediator that may play an important role in growth and survival of carcinomas. In this study, LPA production and response were characterized in two human prostate cancer (CaP) cell lines: PC-3 and Du145.
