The approximate extraction workflow time with the KingFisher Flex instrument is <90 min for 24 DBS samples and <60 min for 24 plasma samples. With the KingFisher Duo instrument, the approximate extraction workflow time is <75 min for 12 DBS samples and <45 min for 12 plasma samples.

The MagMAX Viral/Pathogen Nucleic Acid Isolation Kit, for HIV-1 dried blood spots (DBS) was validated using 200 µL of plasma sample. However, the standard protocol mentioned in the product manual uses reagent volumes that can work with 200-400 µL of sample.

The DBS lysate volume is variable because the filter paper material absorbs anywhere from 150-250 µL of the starting 600 µL of Dried Blood Spots Lysis Solution during incubation. If two 13 mm diameter cutouts are used instead of one, the DBS lysate volume recovered may be even less.

The MagMAX Viral/Pathogen Nucleic Acid Isolation Kit, for HIV-1 dried blood spots (DBS) (Cat. No. A53770) was validated using 1.5 mL non-stick tubes per 13 mm diameter DBS cutout. However, the 1.5 mL tubes have a narrow opening which requires folding the 13 mm DBS cutout into a “taco” shape prior to inserting into the tube. Other tubes (eg., Sarstedt 8 mL) have been used during development and work well, but have more dead space which may result in more sample evaporation.
