The FreeStyle™ 293-F cell line is derived from the 293 cell line and is intended for use with the FreeStyle™ MAX 293 Expression System or FreeStyle™ 293 Expression System. FreeStyle™ 293-F cells are adapted to suspension culture in FreeStyle™ 293 Expression Medium. Frozen cells are supplied in and may be thawed directly into FreeStyle™ 293 Expression Medium. The FreeStyle™ 293-F cell line is supplied in a vial containing 1 mL of cells at 1 x 107 viable cells/mL in 90% FreeStyle™ 293 Expression Medium and 10% DMSO. Characteristics of FreeStyle™ 293-F cells:

• Prepared from low-passage Master Cell Bank cultures derived from parental 293-F cells that were re-cloned by limiting dilution. The 293 clonal derived cultures are maintained in serum-free conditions for only 30 to 35 total passages.
• Adapted to high-density, serum-free, suspension growth and may be maintained in FreeStyle™ 293 Expression Medium.
• Demonstrate high transfection efficiencies with FreeStyle™ MAX reagent.
• Suspension cultures may be transfected in FreeStyle™ 293 Expression Medium without the need to change media.
• Permits transfection of cells at large volumes.
For Research Use Only. Not for use in diagnostic procedures.

Frequently asked questions (FAQs)

Other 293 cell lines may be used with the FreeStyle 293 Expression System. However, they would have to be adapted to serum-free suspension culture in FreeStyle 293 Expression Medium and evaluated for transfection and expression.

When comparing FreeStyle 293-F cells with FreeStyle CHO-S cells for average range of antibody and protein yield, it is definitely expected that antibody yield will be higher in FreeStyle 293-F cells than in FreeStyle CHO-S cells, when generated by transient transfection. On the other hand, antibody yields in stable CHO cells can be much higher. For non-antibody proteins, it is impossible to predict which of the two cell lines may provide higher yields.

You should be able to use the ExpiFectamine293 Reagent for transfection of Freestyle 293-F cells grown in FreeStyle 293 Expression Medium; however, the enhancers will provide only a little boost in expression as they are designed to work with higher density cultures that FreeStyle 293 Expression Medium cannot support.

We typically keep the culture going for 7-10 days posttransfection. After that, protein expression may drop.

Our recommendation of 8% CO2 for the optimal growth of Freestyle 293 cells in Freestyle 293 Expression Medium is based on the bicarbonate content in the media and growth conditions of the cells observed by R&D. That said, you may use lower concentrations of CO2 if necessary (5% is common) to grow these cells. You may observe slightly slower growth kinetics or lower cell concentrations, but functionally the entire system will still work.

Abstract
Molecular dissection of the alpha-dystroglycan- and integrin-binding sites within the globular domain of human laminin-10.

'The adhesive interactions of cells with laminins are mediated by integrins and non-integrin-type receptors such as alpha-dystroglycan and syndecans. Laminins bind to these receptors at the C-terminal globular domain of their alpha chains, but the regions recognized by these receptors have not been mapped precisely. In this study, we sought ... More
Biological activity of follistatin isoforms and follistatin-like-3 is dependent on differential cell surface binding and specificity for activin, myostatin, and bone morphogenetic proteins.

Journal:Endocrinology
PubMed ID:16627583
Follistatin (FST) and FST-like-3 (FSTL3) are activin-binding and neutralization proteins that also bind myostatin. Three FST isoforms have been described that differ in tissue distribution and cell-surface binding activity, suggesting that the FST isoforms and FSTL3 may have some nonoverlapping biological actions. We produced recombinant FST isoforms and FSTL3 and ... More
Mutation of key residues of RPE65 abolishes its enzymatic role as isomerohydrolase in the visual cycle.
Authors:Redmond TM, Poliakov E, Yu S, Tsai JY, Lu Z, Gentleman S,
Journal:Proc Natl Acad Sci U S A
PubMed ID:16150724
RPE65 is essential for isomerization of vitamin A to the visual chromophore. Mutations in RPE65 cause early-onset blindness, and Rpe65-deficient mice lack 11-cis-retinal but overaccumulate alltrans-retinyl esters in the retinal pigment epithelium (RPE). RPE65 is proposed to be a substrate chaperone but may have an enzymatic role because it is ... More
